Nanocrystals for Improving Oral Bioavailability of Drugs: Intestinal Transport Mechanisms and Influencing Factors
نویسندگان
چکیده
With the limitation of solubility and dissolution rate insoluble drugs, following oral administration, they would rifely prove poor volatile bioavailability, which may fail to realize its therapeutic value. The drug nanocrystals are perceived as effective tactic for administration drugs attributes possess many prominent properties such elevating saturation solubility, high loading capacity, improving bioavailability. Based on these advantages, application in delivery has acquired significant achievement, so far more than 20 products have been confirmed market. However, absorption is still facing huge challenges due factors. Intrinsic complex physiological environment intestinal tract two most important factors affecting bioavailability drugs. In addition, research multi-aspect mechanisms promoting gastrointestinal gradually deepened. this review, we summarized recent advances delivered orally, provided an overview progress crossing transport by some new techniques. Meanwhile, relevant were also elaborated detail.

Intestinal Absorption and Factors Influencing Bioavailability of Magnesium-An Update
Background Information on the bioavailability of the essential mineral Mg2+ is sparse. Objective/Method Evaluation of the present knowledge on factors influencing the bioavailability and intestinal absorption of Mg2+. Results Mg2+ is absorbed via a paracellular passive and a transcellular active pathway that involves TRPM6/7 channel proteins. The bioavailability of Mg2+ varies within a broa...
متن کاملFenofibrate Nanocrystals Embedded in Oral Strip-Films for Bioavailability Enhancement
The aim of the present study was to make a fenofibrate (FNB) nanocrystal (NC) by wet media milling, characterizations and formulates into oral strip-films (OSFs). Mechanical properties, redispersion study, and solid-state characterizations results suggested that reduction of drug crystal size at nanoscale and incorporation into OSFs does not affect the solid-state properties of the drug. In vit...
متن کاملOral Absorption, Intestinal Metabolism and Human Oral Bioavailability
Most of the drugs that are available in the marketplace are administered via the oral route, which is a convenient and cost effective route of administration (Lipinski 1995; Lipinski 2000; Lipinski et al. 2001; Lipinski 2004; Abrahamsson and Lennernas 2005). Thus, oral bioavailability is one of the key considerations for discovery and development of a new chemical entity (NCE). It is well recog...
متن کاملImproving intestinal absorption and oral bioavailability of curcumin via taurocholic acid-modified nanostructured lipid carriers
The expression of multiple receptors on intestinal epithelial cells enables an actively targeted carrier to significantly enhance the oral delivery of payloads. Conjugating the receptors' ligands on the surfaces of a particulate-delivery system allows site-specific targeting. Here, we used taurocholic acid (TCA) as a ligand for uptake of nanostructured lipid carriers (NLCs) mediated by a bile-a...
متن کاملFactors Influencing Oral Bioavailability of Thai Mango Seed Kernel Extract and Its Key Phenolic Principles.
Mango seed kernel extract (MSKE) and its key components (gallic acid, GA; methyl gallate, MG; and pentagalloyl glucopyranose, PGG) have generated interest because of their pharmacological activities. To develop the potential use of the key components in MSKE as natural therapeutic agents, their pharmacokinetic data are necessary. Therefore, this study was performed to evaluate the factors affec...
